Distributional Changes of Neurofilament 68 kDa in the Cerebral Cortex following Transient Gerbil Forebrain Ischemia

Cerebral ischemia produces extensive neuronal degeneration in several regions of the forebrain. Neurofilament (NF) proteins not only support neurons as cytoskeletal components but also play a fundamental role in axonal transport. In order to investigate the correlation between NF and neuronal degeneration, the distribution of NF proteins was studied immunohistochemically in the ischemic cerebral cortex with antibodies against NF 68 kDa protein.
In the sham operated group, NF 68 kDa immunoreactivity was distributed in cell bodies and main processes in layers III and VI of the neocortex. At 1 day after ischemia-reperfusion, strong NF 68 kDa immunoreactivity markedly increased in cell bodies and processes in the layers III and VI. NF 68 kDa immunoreactivity at 2-4 days after ischemia-reperfusion was weaker than that at 1 day. These findings suggest that the early increase of NF 68 kDa in layers III and VI of the neocortex after ischemia may be related closely to the selective vulnerability of these particular regions in case of ischemia.
